چکیده

1. Role of c-AMP: Calcitonin binds to specific calcitonin receptors on the plasma membrane of bone osteoclasts and renal tubular epithelial cells, activates adenyl cyclase which increase c-AMP level which mediates the cellular effect of the hormone. This is the principal mechanism by which calcitonin acts. 2. Celluar Shift: It has been suggested that calcitonin may directly affect the relative distribution of bone cells. The hormone both in vitro and in vivo produced a celluar shift, in which the number of osteoclasts decreased. 3. PH change: Calcitonin may regulate PH at cellular level producing more alkaline medium which diminishes resorption. METABOLIC ROLE

Selective internalization of the apical plasma membrane and rapid redistribution of lysosomal enzymes and mannose 6-phosphate receptors during osteoclast inactivation by calcitonin.

The effects of inhibition of bone resorption by the peptide hormone calcitonin have been studied at the level of the osteoclast. Although not epithelial, the osteoclast is polarized with the secretion of newly synthesized lysosomal enzymes and of acid occurring specifically at the apical pole, facing the bone compartment. The membranes composing the apical (ruffled-border) and basolateral domai...

Calcitonin gene-related peptide receptor antagonist human CGRP-(8-37).

From this study, we predicted that the human calcitonin gene-related peptide (hCGRP) fragment hCGRP-(8-37) would be a selective antagonist for CGRP receptors but an agonist for calcitonin (CT) receptors. In rat liver plasma membrane, where CGRP receptors predominate and CT appears to act through these receptors, hCGRP-(8-37) dose dependently displaced 125I-[Tyr0]rat CGRP binding. However, hCGRP...

Abundant Calcitonin Receptors in Isolated Rat Osteoclasts

Calcitonin receptors have been characterized for the first time in isolated osteoclasts. These receptors have been demonstrated by autoradiographic and biochemical methods, and the cells have also been shown to respond to calcitonin with a dose-dependent increase in cyclic AMP. The receptors in rat osteoclasts are specific and of high affinity (dissociation constant, Kd, 1 to 6 X 10-10 M), and ...

Amylin increases cyclic AMP formation in L6 myocytes through calcitonin gene-related peptide receptors.

The cellular function of amylin is investigated in L6 myocytes, a rat skeletal muscle cell line. Both rat amylin and human amylin-amide acutely cause a dose-dependent increase in cyclic AMP formation in L6 myocytes. 100 nM amylin stimulates intracellular cyclic AMP concentrations 12-fold, whereas human amylin-amide at this concentration causes only a 2-fold increase. Up to 10 mM human amylin ha...
